I am using inducible promoters to express ß-carotene in yeast S. cerevisiae cell. Yeast ß-carotene producing colonies are able to show orange color(light to dark). I transferred different promoter with variety of strength in term of transcriptional output. we hope to see different colorful colonies referring to affect of different promoter in single plate. Now the problem is how I can express promoters or how I can produce ß-carotene on solid media.

IPTG can't be used straight forward as an inducer in yeast because there is no natural lac promoter in yeast. However some recent data prove that IPTG regulated promoter may be constructed in fission yeast. For more info:

There is absolutely no problem: this is the basis of alpha complementation on agar media plus X-Gal/IPTG (blue/white clone screening). Provided the coloration due to carotene becomes visible.
