I've tried IFN-gamma at 200 IU/ml but not seen any upregulation in surface MHC class I in Vero cells. This concentration typically gives me a 3-fold increase in surface expression in porcine primary macrophages.
I was thinking about trying TNF-alpha or a combination of TNF-alpha and IFN-gamma. Maybe even IFN-alpha.
Has anyone managed to stimulate MHC class I in Vero cells, and if so, how did you do it?

I am not familiar with the Vero cells but in our lab the IFN activation can take up to 3 days. Anyway every cell line requires its own calibration. Besides that there are some cell lines which do not respond to IFN may be due to the lack in the receptor to IFN-gamna
