During our research, TBARS assay was carried out for cheese samples on the basis of the Standard Method of AOCS (Official Method Cd 19–90). Briefly, 50 mg of each cheese sample was weighed in a 25 mL volumetric flask, and its volume adjusted with 1-butanol. Then, 5 mL of the TBA reagent was added to 5 mL of the test solution and placed into a thermostatic bath at 95 °C After 120 min the test tube was removed from the bath and cooled under running tap water for about 10 min until its temperature reached to 20 °C. Then a 10 mm cuvette was used to read the absorbance of the reaction solution at 530 nm. I want to know what should we do for the standard curve of MDA? I mean, should we prepare different concentrations of MDA in butanol?
