Let suppose we make a PDMS lines( 50um) on glass/PET substrate and after fully curing PDMS, we cover the PDMS lines by spin coating with PDMS and again cured the whole sample. Now, how can I measure the adhesion/ force of PDMS lines with substrate and coated PDMS? Also what will be the effect of PDMS width on adhesion? PDMS (10:1)

Note, that this top-down method gave one of the highest values of that energy ever published (20% higher than my own bottom-up method value btw), but you should get into a proper range.
You can use force curves to calculate adhesion between two materials .... one of the materials must coated on the tip of AFM device , then you can use force curves to measure adhesion between two different material.

You can use the force to test the adhesion between two stacked layers. One can use adhesive film and pull it with a a force sufficient to separate the two layers.
Another method may be an optical one:
If the layers are identical in construction and made to join each other then if they are chemically bonded there will be no gaps between the layers.
If subjected to light very little light will be reflected from the interfaces if the layers are good bonded to each other and if they are not good bonded light will be reflected from the interface and in these way one can use the reflected light as an indication of the quality of the bond.
