I have extracted plant leaves in crude form I did not went for any vaporization or deep freezing now. I would like to know antioxident activity and phenolic group concentration, how can I know concentration of phenolic groups in plant extract?

Use Folin Ciocalteu method to determine phenolics. Gallic acid can be used as standard. For antioxidant activity, you can use DPPH, NBT, ABTS, FRAP etc. I am attaching a paper for your perusal. I think it would be a perfect reference for your work.

By using Folin Ciocalteu method you can determine total phenolic compounds , but you can use another standard compound like catechin or caffeic acid. You can also measure the amount of orthodiphenol by using sodium molibdate solution 5% dissolved in 50% ethanol and, then the absorption will be measured after 15 min at 370 nm by a UV-visible spectrophotometer. To know the flavonoids content, use aluminum chloride colorimetric method.

First try to know the concentration of extract for dilution. As you work with some weighed quantity of raw material with specific quantity of solvent. Try to find out the concentration of extract in w/v. Then try above method.
