I would like to isolate flagella from Leishmania parasites. There are numerous publications that do so, however by procedures that involve detergent and salt extractions to depolymerize the subpellicular microtubules; leaving the flagellum intact. However, I am interested in studying axonemal/flagellar dyneins of Leishmania and was wondering that such high salt extraction might also extract out the flagellar dyneins.
Are there any protocols that can be used to isolate Leishmania flagella, in similar ways which are applied to Chlamydomonas and other organisms?? (eg. pH shock, dibucaine etc).
