My spinoculation protocol provides high titers of of virus at 24 and 48 hours by p24 ELISA. However, my proviral DNA qPCR data as well as p24 flow cytometry suggests that only ~5% of the cells are infected at 24 hours. I'd like to know some ways to increase the number of infected cells in culture within 24 to 48 hours without increasing my MOI? Thanks.
