I've prepared nanoemulsion and need to measure the release and permeation of that formulation using franz diffuiosn cell. I've found some of the researchers added SDS (sodium dodecyl sulphate, 2%) or ethanol in PBS 7.4? Which one is the best? Is there any guideline that I can follow for this experiment? My drug is very hydrophobic (0.039 ng/ml - solubility in water) and high solubility in methanol and ethanol. Thanks a lot.
Increasing the temperature may work; however, if its a transport experiment this may affect your results. You could also play with the the pH this may help as well.
If surfactants are the way you want to go, adding DMF or DMSO is another good route.
Solubility of hydrophobic drugs can be increased by conjugating drug with different excipients like hypromellose, povidone, sodium lauryl sulfate, PEG, p-(meth)acrylates (e.g. Eudragit E100), Kollicoat IR, TPGS etc using different techniques of solid dispersion
Solubility of a hydrophobic drug can also be enhanced by complexation with cyclodextrins or by by mechanochemical activation technique using various hydrophillic carriers such as lactose etc., which avoid various organic solvents..
You can used pH dependent solubility of drug molecules. If your drug has higher solubility at lower pH than you can used that. Let's say your drug is more soluble at pH 4.5 than you can used that pH buffer with 10% ethanol. Moreover, replace whole media more frequently.
A use of a cosolvent in the acceptor compartment (like glycerol or propylene glycol) might help. Cyclodextrins too may complex your drug and increase its solubility. You have to determine the solubility of your drug at the temperature of the experiment with the new conditions and dose accordingly your active in the donor to keep sink conditions.

In my opinion you are developing formulation for the topical route. Skin pH ranges 4 to 5.5. You can use phosphate or acetate buffer pH 5 and 0.05% sodium lauryl sulphate. I think it might be beneficial for you in your study.
