How can I increase protein yield from insect cells?

Hi all.

I'm trying to express protein from insect cell culture with baculovirus expression system. This is nothing new, there are already lots of publications saying that is easy!!!

But I am getting too low proteins from insect cells.

I'm using baculovirus system from invitrogen and Sf9 cells.

Donor plasmid was sequenced and everything is ok.

I'm quantifying expressed protein by ELISA and getting reaction, that is suggesting my virus is infecting and expressing proteins.

But I have no idea why proteins is too low.

Please anyone has suggestion, let me know!

Hola Sungseok, If you want to have high yield of recombinant proteins, you have to see the majority of your cells your cells with symtoms of infection , detached, floating( if you work with monolayer) and swollen. I supossed that you have made diferent passages of virus (p1,p2,p3)and the cells are infected whit high (3-10) multiplicity of infection.. if no, you have to let growing the cells more time untill you can see that symptoms. good day and good luck
