I have different plates with different strains. The next step consists of doing a suspension of each microorganism. I do not have any problem with E. coli, S. aureus, etc, but I do with B. subtilis. I normally use Ringer solution, PBS pH 7,2 o similar buffers to do the suspensión. I pick up a colony or colonies and inoculate the buffer. Most of the strains dissolve in the buffer but B. subilitis doesn't. why? Which buffer or culture could be proper to dissolve Bacillus and get its suspension?
