I can suggest an efficient method describe by Della Porta et al., 1983 Plant Mol. Biol. Rep. 1, pp. 19-21 for plant DNA miniprep but useful also for nematodes.
I am doing PCR on a COX II gene on a nematode using the followings primers
COXII F TCT AGT TAT ATA GAT TGR TTT YAT
COXII R CAC CAA CTC TTA AAA TTA TC
It is mention to cite I have to use this primers to get a 629 bp Band the problem
that I face back to the primers as my PCR just work well for less than 10 days and after that period I fail to run a successful PCR . How can I elevate the life of primers I recently change my company an ordered a new one but unfortunately the problem has persist to me yet. I work with ITS related primers in parallel but never faced such problem.
PS: I used DNA free water to wet my primers as company ordered.
