I performed a tissue allogenic transplantation, and I want to determine that whether the endogeneous cells are viable post transplantation. Now, the tissue has been fixed and embeded in paraffin. So, is there some staining that I can perform to research my goal? Thank you.
Check upon staining with Jenus green dye without fixing or embedding the retreived cells under light microsocope.
You can just perform a simple pathology evaluation. stain the tissue sections with hematoxilin and eosin and evaluate the cell morphology, eg shape and size of nucleus, citoplasm color, presence of blebbing or necrosis of cells, and so forth.
The suggestion of labelling your cells is very good, after all you have to prove that they are what you say they are in the tissue section, unless the morphology is clearly different from the tissue.
