I do not have any experience with KG-1 suspension cell culture. Do the cells require agitation or can I culture them in simple T-75 flasks? What is the best way to culture them?
I've carried KG-1 for about a decade. It is a relatively simple cell line to culture. I use IMDM + 20% FBS with antibiotics as the culture medium. I have carried it in T25 and T75 flasks in %5 O2/5% CO2, but low-ox is not necessary for KG-1. I split it once a week 1:5 then feed 1:3 four days later. In low-ox, it can reach >2.5 x 10^6 cells per milliliter.
I agree with Gary Lee Gilmore. Do not centrifuge the cells for passaging, use the splitting method, which explained above. Good luck.

In addition, I would say IMDM is better than RPMI 1640. You can supplement it with 20%FBS as mentioned by Dr. Gilmore and 2mM L-glutamine. Seeding density is important. Do not culture below 1 x 10^5 viable cells per ml.
