I am working on CD133+ cancer stem cells from B16F10 cell line. I use serum-free high glukose DMEM medium and also add EGF, EGF as growth factor. But CD133+ cells died after 3 days. Do you have a any suggestion for culturing CD133+ cells? I couldn’t find which media is best for cancer stem cell medium.

Hello, We culture breast cancer stem cell from SUM159 and MDA-MB-231 cell line with serum-free DMEM/F12 media (1:1), B27 (1:50), EGF (20ng/ml) and bFGF (20ng/ml), insulin (5μg/ml), hydrocortisone (5μg/ml). In general, at least in the case for glioblastoma cells, it is important to use serum-free medium compositions, as FBS usually leads to a differentiation of CSCs. Nevertheless, culturing a pure CD133 positive population is, at least in the case of glioblastoma cells, impossible, as the population has shows a kind of plasticity, meaning CD133+ cells can give rise to CD133- cells, so you rather enrich for positive cells, but you can not propagate a purely positive population. As youmentin that your cells die after 3 days, how are you sorting for your positive population (FACS, MACS, ...)? Is there a chance that the cells just do not survive the sorting procedure (have you checked viability after the sort)? In that case it might be of help to improve the sorting conditions to ensure cell survival after the sorting.- Badges
