I isolated CD8 T cells from murine spleen and thymus, then check their chemotaxis ability using Boyden chamber assay. I counted the number of migrated cells in lower chamber by hemacytometer. However, I think this method is not really accurate. And in some papers I read, they used flow cytometry. Can anyone tell me to quantify Cd8-positive cells by FACS ? I appreciate your help. Thank you
