I’ve performed flow cytometry of peripheral blood leukocytes from mice to identify and quantify inflammatory and non-inflammatory monocytes. I have marked my cells with LIN, CD11b, CCR2, CX3CR1 and GR-1 to be able to differentiate both types of monocytes but I’m finding it hard to establish the gates properly in the analysis software (Kaluza). I started identifying monocytes with a LIN-CD11b plot, but after that I don’t know what plots and gates are going to be the best. I’ve tried with a CCR2-GR-1 plot, which seems to be fine for identifying both types of monocytes, but I wanted to somehow include CX3CR1 to the analysis too. Can you recommend a better way to identify them or how can I improve my analysis?
