you have to determine the mass concentration (in mg/mL) and then use it to divide activity in U/mL.

If I understand your question correctly, then you have an enzyme activity in U/(mL enzyme formulation) and a protein concentration in (mg enzyme protein)/(g enzyme formulation), and you want to know the specific activity in U/(mg enzyme protein)? Assuming that the density of your liquid enzyme formulation is approx 1, you just divide U
In my point of view you need to freeze dry the solution or with other mild drying method which doesn't influence significantly the enzymatic activity. And then run your enzymatic activity assay with predetermined weight of your enzyme.
To get specific activity u/mg caluculate per mg protein from u/ml.
