When reporting the IC50 values of a curve dose-response obtained with Prism-(Non linear regression, log conc-normalized response), do I need to report the SEM? If yes, can I calculate it from GraphPAD or should I calculate it with Excel? Graphpad gives the LogIC50 and there is a math reason for that (I am not quite sure I understood though).

For each sample (repeat) you should calculate IC50 and from calculated values you can further extract the SEM (in Excel where its simpler for me at least).
Hi - you can use GraphPad to calculate if you want to. Click:
Analyze
XY analyses
Row means/totals
Check the "Row means with SEM" circle
Click OK.
Use new table and graph==> XY
Put how many replicate of data in Y list, for example, 3 or 6, depend on number of data you have.
After that analysis==>normalize==>non linear regretion==>inhibitor vs normalized response-variable slope
after that you can get the IC50 data and its SD as well..good luck!
