Is it possible to do Central composite design and Response surface method when there are two independent variables? Say, the first independent variable represents 5 graded levels of a fatty acid and the second independent variable represents 2 temperature and I would like to see their interaction effect on fish.
Please provide me suggestion how it can be analysed in Excel or SPSS.

Question: Are the 5 levels in the first variable necessary? Or can they be reduced to 2?
The response surface implies that you have an equation that allows you to describe the results in a spatial region (your study region). My recommendation is: start with a full Factorial Design, over a wide range, and then see if the axial and central points of the composite central design are required. Having said that, I think that in Excel it would be easier for you to implement the calculation of the following experimental steps.
