How can CRISPR-mediated targeted knock-in achieve gene expression levels more than ten times higher than the baseline? Literature reports that lentiviral infection followed by screening for high-copy clones can achieve this goal. Are there any specific expression strategies in targeted knock-in that can achieve similarly high expression levels? The currently used EF1α promoter cannot reach such high levels when expressed at conventional safe harbor sites.
