I am working on the gene expression of Passion fruit wax biosynthesis.
I have selected the sequence of the primers (forward and reverse) from a published article about Passion fruit. but if I want to make sure whether those selected primers sequences exist in the passion fruit genome and are specific to Passion fruit or not, so how can check?
Use BLAST and run against the passion fruit genome to make sure the primers are in passion fruit.
Why do you need to know if the primers are specific to just passion fruit? That's harder to answer unless the gene in question is KNOWN to be unique to just passion fruit. are you dealing with samples that have multiple DNA sources?

As I did the q-PCR by using the primers from the article and they are not expressing with the same conditions, (even the same variety), so that's why, I was want to make sure that weather those primer sequences are specific to passion fruit or designed by using the other references
The easiest way is to use Primer-BLAST (https://www.ncbi.nlm.nih.gov/tools/primer-blast/index.cgi?LINK_LOC=BlastHome ). Use your own primers and enter an organism name (for example Passiflora (taxid:3684)) in Primer Pair Specificity Checking Parameters
