When I defrost my plasma samples from -80, I do so on ice, slowly. Then I centrifuge the samples. Upon centrifugation, the plasma samples will have a white sticky substance layer at the top, but sometimes throughout. I avoid this with my tip as best as possible.

My results were anything but consistent. I am wondering should I vortex to force the sticky substance into solution? Does this substance, likely precipitate contain the cytokines of interest and that could explain my results?

Some of my plasma samples are over 10 years old, but have never been thawed before. SO I get how the inconsistencies could be the age of the samples.
