I need a suggestion to isolate animal DNA from tallow or beef fat..
Proceesed tallow doesnt allow recovery of DNA as it has been boiled over 110 deg celcius. Still i need a method to work with a spike or postive control?
How to separate cells from fat...i have read some literature regarding centrifuge after treating fat with n-Hexane or PBS or CTAB buffer....can any of it help...?

Why do you need to separate cells from fat? Would it be possble to just chop or homogenize your fat, and then perform a standard phenol chloroform precipitation method?
Arseniy .. My sample is actually processed fat avaialable as tallow. And it seems my traeitional ctab method is not working perfect...i want some suggestion which i can try on spiked tallow..
I have checked the method. We should check the critical point. As the samples is high fat content, should be careful when separate fat and DNA. The tallow is high content of triglycerides than cells, so the amount of DNA maybe lower compared to another tissues.
Grinding samples after adding liquid nitrogen may improve your method
The CTAB is good to use when extract DNA in this matrix, however I recommend you to use sodium dueodecil sulfate (SDS) as alternative. We have face this problem when extract DNA in high fat content samples.
You may try to increase the amount of CTAB.
