It is good that you have DMEM. But it would have been still better if you would have also had Ham’s F-12 which perhaps may not be that expensive.
Use the two media in the ratio of 1:1. You may add 10% FBS as supplement to this mixture. There are other supplements which may be required such as 2mM L-glutamine, 1mM sodium pyruvate and 400 ng/ml hydrocortisone (maintains proliferation at a slightly greater rate). Hope you may be having these supplements in the lab.
The above recipe would be much cheaper than Keratinocyte Growth Medium(KGM). But optimization of this lab-made media might be required, especially modifying the ratio of the two media. You may also try DMEM: Ham's F-12 in the ratio 3:1.
Hi Malcom, I would like to follow up on the previous question as I found that some researchers have been successfully cultured primary gingival keratinocytes using DMEM/F12 in the ratio 3:1, as you have previously mentioned. However, I am not sure to which DMEM were you referring to? High or low glucose?
