Hello Sri Charitha , unfortunately I have never cultivated RL95-2 cells, so my answer will not help you on the specifics of their culture (medium, etc).
However, in the facility where I work, we regularly receive vials from ATCC and the quality of the frozen cells is usually deplorable ... They require some patience and some extra care during the first days / weeks. It is a shame in my opinion, but also not surprising in my experience : I would not worry too much about whether the conditions I use are the best. 24 hours is really soon, last time when we received ATCC cells, we thawed the vial on a wednesday in a T-25cm² and the cells were not properly attached and good-looking until the following monday ...
But if the cells are in too bad a shape and do not seem to get better, I would not hesitate to ask that they send a new vial : I think you only have a month after you receive the cells for making a complaint ?
