The first things to to is to check whether or not the TP53 gene is mutated
Then, you can check how your cell line behave after DNA damage (G1 arrest, apoptosis)
There are a lot of assay depending on the initial question
Hi Maria - perhaps you can be more specific?
Are you wanting to know about p53 in different cell types? (ie cancer and "normal").
In general, you will need to be able to do several techniques to investigate this thoroughly (such as western blot, elisa, qPCR etc...)
Lentiviral silencing of the TP53 gene would also be interesting to see changes downstream.
I would recommend getting a specific question in mind that you want to answer and go from there.

First, determine mutation status in your cell lines.
DNA damaging agents can be used to induce p53 , its stability/ protein levels should go up.
Also look at target genes via qPRC and WB ; p21, PUMA , NOXA
