Hi, everyone. I need to perform a real time reverse-transcription PCR in one step. What we have is SYBR green I PCR Master mix with GoTaq DNA Polymerase. So far, our attemps to perform the assay in one step with this reagent have been unsuccessful. Have anyone tried this successfully? What do you recomend?

Dear Yulaimy,
You said above one step qRT-PCR with Go Taq polymerase. I suggest you to produce your cDNA in a first reaction with a Transcriptase Reverse enzyme. Thus, you can use your cDNA in a second reaction with Taq Polymerase and SYBR green. Good luck!
