I would like to express a protein from a pAAV but using direct transfection of the plasmid in the cell lines of interest. It should be possible since i saw papers that did it but does anyone already try it and could tell me if this is really efficient for protein expression?

Yes, you can use the AAV plasmid. It will work as well, if not better, then AAV. It doesn't matter how you get the expression cassette into the cell (plasmid vs AAV).
We always transfect our AAV plasmids before making AAV batches to confirm that the expression cassette is functional.

@Jonathan - do you perform the transfections and further culturing under higher safety conditions or as usual? I am just confused whether AAV belong to the normal or higher safety level..
