I am working on HMC3 cells and I see there is mixed responses on using LPS and ATP for IL-6 detection. I understand we use this LPS +ATP for release of IL-1β and IL-18. But I do know that IL-1β can be used to induce other cytokines. so, I would like to know if anyone has used LPS and ATP for IL-6 detection

If I understand correctly, you primed the inflammasome in your monocytic cell line using LPS+ATP and then measured IL-1b, IL-18 and IL-6. LPS induces all kind of cytokines including IL-6 (lots of it). So it is not a surprise to see IL-6 here. In fact, adding ATP activates inflammasome so that you get IL-1/IL-18 cleavage/release on top of the inflammatory response that is induced by LPS (which includes a variety of cytokines/chemokines).

I have used LPS+ATP in HMC-3 cells to activate the NLRP3 signaling cascade. LPS-1ug/ml for 6-8hrs followed by ATP-5mM for 1hr was sufficient to show marked difference in the production of IL-1B, IL-18, TNF-a and IL-6.
* I measured cytokines using BD bead based cytokine assay kit.
