I used 5x106 cells injected with matrigel into the 4th mammary fat pad of Balb/c nude mice. The mice formed tumors, however none of them reached 100mm3. I have read several papers stating the same protocol and their tumors reached 100mm3 in 3-4 weeks. My tumors reached about 50 in 4-5 weeks and then started getting smaller. Thank you in advance for any help with this!
You may not be doing anything wrong.
The number of cells you are using is more than enough.
The problem maybe the way different labs measure tumor size.
Just start your treatments early enough to see the effects.
looks like the problem is with mice model, try different supplier or different model.
Try using cells with lesser sub-passages.

We've done breast cancer xenografts commercially (http://altogenlabs.com/xenograft-models/breast-cancer-xenograft/) and you should be on track using NOD/SCID mice with original cell lines. I would recommend using matrigel during the initial injection, as it helps keep track of the tumor size and forms a coherent lump from the start.
