I've tried inducing active systemic anaphylaxis based on a paper. Briefly, rats were sensitised with 10 microg of OVA + 10 mg of alum on Day 1 followed by induction with 1 mg of OVA on Day 14. Through observation, the induced rats showed signs of scratching. However, I couldn't detect TNF-alpha and IL-4 from the serum collected using commercial ELISA kits. A few possibilities on why it didn't work were suggested - dosage, the number of sensitisation, and ELISA may not be the best detection method. Hence, I need help on establishing a workable model.
