In the Renca renal cell tumor model, has anyone observed deaths in control BALB/c mice with moderate tumors (~500 mm^3)? Mice appear to be healthy with no body weight loss but are found dead the next day. Could this be a result of the production of a tumor related factor such as IL-6 or TGF-b? I have asked other investigators using the Renca model and they have not observed this sudden death.
Have you done necropsies on these mice? We have not seen sudden death in our Renca line but have seen it with other syngeneic lines and found a severe metastisis to the lungs or abdomial cavity adjacent to the area of cell implant.
Renca is known to be an aggressively-growing cell line, so it's no surprise that you could be getting metastasis. Compare your protocol with standard ones for Renca xenografts (see http://altogenlabs.com/xenograft-models/kidney-cancer-xenograft/renca-xenograft-model/) and that should help you find out any potential causes unexplained by standard error. Sudden death can occur due to a number of reasons, and necropsies can definitely help pinpoint the issue.
