I use Cyaman Chm ELISA kits (https://www.caymanchem.com/app/template/Product.vm/catalog/582701). The testosterone kit has shown to be unreliable. The results have not been reproducible. A large kit works only the first time, but then within a few days it stops working. Specifically, either the standard curve falls apart, the assay does not develop well, the samples come up with very high background, etc. In short, the problems are not consistent. I would love to hear from someone who has recently run this kit successfully. Thanks

Yes the mentioned problem is exist for Cayman Tes after the first run. I suggest you Diagnostic Biochem Canada (dbc ELIA for Tes). The enzyme is HRP and it has very good stability and acceptable precision.

Kits are designed more for industrial or clinical use and is meant to be thawed, diluted, etc and used up in one assay day.

I had exactly the same problem, even worse, it did not work at all (high back ground noise). I wasted considerable amount of money and time.
I am using Hospitals' Biochemistry Laboratory system (Hitachi E170 auto analyser- based on electrochemical chemoluminance method- according to the protocol provided by the manufacturer. The assay range was 0.06–0.82 ng ml−1 ). I did not have any problem so far, except high reading from castrated rats.
I guess that is nothing to do with reading system, simply testosterone produced by adrenal glands.

Just new on RG and found your post. This might not be any more of interest for you, but we have developed the technology used in Cayman kits. Should you be interested to get information and assistance, feel free to get in contact. HRP and AcHE are quite different, but the problem comes more probably from water. We have experience this for several customer and helped them troubleshoot.
