Could somebody tell me if the using of the Nanodrop 2000 could be good to measure Abeta concentration after oligomerization in a sample prepared using DMSO and Neurobasal?
I use NanoDrop to check that I have reconstituted my peptide AB40 properly. My protocol is as follows
1)Open NanoDrop
2) Select Protien 280
3) Change Type to "Other Protien (EMW)
4) Enter (1.49) in sigma/1000 *Literature Value
5) Enter (4.33) in MW (kDa)
6) Type in Sample ID
7) Click Blank
8) Pipette 1.5uL of sample onto the NanoDrop
9) Click Measure
10) Repeat step 9 till concentration is constant
However I don't find NanoDrop as helpful when it comes to my buffer system. I too am looking for a good protocol for this as well. I'm told because I reconstitute my peptide with 60mM NaOH NanoDrop this is probably not as accurate as I'd like to believe it is.

Could I ask which kind of amyloid do you use and how do you prepare your amyloid to be aggregated? I bought amyloid 1-42 and want to use it to build Alzheimer disease model. I don not know how to prepare it and make it have toxic to cell. Thanks....
