Hi,
I am really at a loss here. I am differentiating Thp-1 into macrophages, and then polarizing them into M1 and M2 macrophages. My M1 profile is basically flawless, but I can't get M2s for the life of me. I have tried like, a ton of protocols. I was able to get a super clear CD206 overexpression in M2-polarized blood monocytes (from human PBMCs), but I lost it when switching to cell lines. If anyone has worked on this, I'd really like some help :)
