From what I've read these cells are already Hygromycin-B resistant. I was wondering if it is still possible to select for stably transfected cells using a really high concentration of hygromycin?
I did transfection in OSCC cells and which are found hygromycin B resistance too. As I use double concentration, cells die slowly. However, As I combine hygromycin B and puromycin, stable clone could be quickly selected. You can also try combining hygromycin B and puromycin. The concentration of hygromycin B and puromycin I use are 350 ug/mL and 5 ug/mL, respectively, which is referred to instruction of Lantiviral activation particle from Santa Cruz Biotech.
