I want to fix a Superose 6 10/300 GL column to our AKTA prime system. However, the product info says that the Superose 6 10/300 GL is not suitable for use with the AKTA prime plus system. Does anyone know why? When I try to look up the AKTA prime system I can't find any info from GE, so I'm wondering if this system has been discontinued? Or if it is in fact the same as the AKTA prime plus system? Any pointers much appreciated!
The AKTA Prime Plus replaced the AKTA Prime because the piston pump of the Prime Plus is better than the Diaphragm pump of the Prime in all the various types of pumping demands, maintenance, and other specifications. How you want to do direct loading is a matter of choice.
Neither the old Akta prime, nor the newer Akta Prime plus (in spite of the better pump) will be able to operate at a sufficient pressure to run the superose 6 10/300 at maximum (recommended) flowrate.
This is probably the reason why you got such informations.
You could probably run it at lower flowrate, but the regularity of the flow and the resolution of your run would not be guaranteed.
Thanks to the both of you for your helpful replies. My options are either the akta prime or an akta xpress, which although is also not listed as a compatible system, we can modify in order to attach the column. I'm not sure which one would be preferable?
I would definitely choose the Akta xpress system, even if it is a bit more complicated to set up and designed to work with rather high flowrates, you can be sure that the pumping system is powerfull and precise enough for that kind of columns.
The Akta Prime (diaphragm pump) and Prime Plus (triple piston pump) are both low pressure systems. The piston pump of the Prime Plus is the same mechanism as the sample pump in the larger systems (like Explorer).
The Prime & Prime Plus operate to 1 MPa max, making them suitable for low pressure purifications only, like affinity and ion exchange. In these, higher pressures damage the chromatography medium. These work as starter systems, although the user interface (same on both) is four membrane buttons - the kind you sort of know you pushed, but you're not sure, and the display is a two-line text display. Both Prime products work with PrimeView software, not Unicorn, as the larger Akta systems do. PrimeView essentially allows you to view the chromatogram and related fraction positions.
Akta xpress, according to GE's website: designed for automated, unattended, multi step protein purification of both single and multiple samples. One to four proteins cab be processed in one run.
