I intend to grow monolayer of cells on transwell insert coated with collagen. I want to eventually extract RNA but will these cells invade and enter through the collagen(invasion) making it harder to extract RNA off the surface? Migration through pore wont be an issue as the pore size is only 0.4um.
What I´m concerned with is invasion through collagen.
In my opinion, cells may enter collagen gel, it does not matter, but will remain upto transwell membrane. At the end of your experiment, you should place the whole transwell in RNA later, and with sterile scalpel, cut the round membrane of cells with collagen and cells. Transfer this membrane in Trizol and use RNA isolation after crushing the disc in a hand-driven glass driven homogenizer. We do isolate RNA from cartilage which is collagen containing cells, we do not isolate cells from cartilage's collagen for isolation of RNA. Hope that helps
