So, the problem is that I deleted 2 genes essential for plaque formation (I am talking about Poxvirus - Cowpox virus).I found information that Poxvirus can replicate only in the RK13 but without 2 genes (E3L and K3L) I do not see plaques. Can it work like this? Virus replicates but I cannot see it? As far as I could find, RK13 is the only cell line where it can occur.
Have not worked with poxviruses but plaque formation requires cell death. It is very possible for viral replication and release to occur without cell death. In the absence of plaque formation, a different assay to titer your virus, such as a fluorescent staining assay if you have a suitable antibody or qPCR.
Good Morning! The best cell growth for poxviruses is Vero cell. Use the RK13 cell and, make a PCR or real Time to quantify.
