Can anyone help me with a protocol for extraction and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of proteins from Cordyceps sinensis and Cordyceps militaris?
What the type of fungi protein that you want to analyze with SDS-PAGE?
(Intracellular or Extracelullar proteins)?
The extraction method depend on the type of your target protein. You can extract the extracellular protein from the supernatant of your liquid medium. If you want to extract intracellular protein, you must separated micelium from the media, adding liquid nitrogen and than homogenized. After that, you can use extraction buffer.
For the SDS-PAGE, you can read on the Bio-Rad protocol that was attached. This protocol very complete.

You can read my newest publication for SDS-PAGE analysis, if your target protein has low Molecular weight (10-25 kDa), you can use 12% separating gels.
Those concentration are good for separation low Molecular weight proteins.
