Hi all,

I sectioned fresh frozen brains (CO2 euthanized rats) using a cryostat. I typically do immunohistochemistry with perfused free-floating brains, but I want to try it with fresh frozen brains. The antibody I want to use is S100 from DAKO and I normally use the ready-to-use anti-rabbit IgG (HRP conjugated) and DAB kit from Vector Labs. I know it's hard to do immunohistochemistry with fresh frozen sections, but I was wondering if anyone knew how to do this. Even if you haven't done S100, I would like to know if there's a protocol for it.

Also, I need to stain 160 slides, and I was wondering if you guys have any tips for doing the immunohistochemisty efficiently.

Thank you!!!

Best,

Andrew
