We are going to run an assay in which a Fluorescein-labelled plasmid is spotted on a hydrogel (3% polyacrylamide) coated slides and then imaged. I was wondering how sensitive the final fluorescence will be to dryness of a hydrogel - if the hydrogel gets dry for some time in the course of the experiment, or if it gets dry during imaging - is it going to affect the fluorescence?
