I am planning to DNA promoter methylation in breast cancer tissue, formalin fixed paraffin embedded tissue and breast cancer cell lines. Which bisulfite modification kit will be most effective for qMSP
All I can comment on is that the quality of the metabisulfite is crucial ie fresh and dry. It is required at a huge molar excess (3M I think) so if it is a bit old, it loses activity and just won't work as efficiently. You'd have to judge either kit by reviews/price - sorry can't help more than that - but look after the reagents once they arrive, put the bisulfite (sealed well) in a dessicator if you have one.
Hi, in my experience Zymo kits are better. Particularly, I suggest EZ DNA methylation-direct kit (https://www.zymoresearch.eu/ez-dna-methylation-direct-kit). It allows you DNA bisulfite consversion directly from your cell/tissue sample avoiding prior DNA purification.
