I am extracting proteins from plant leaf samples by using Tris EDTA method but the problem is I am getting protein but on running sample on SDS PAGE of 15% gel I m not getting clear bands either I m finding light one or two band or bands with smear can I know how to overcome this problem for characterization of antimicrobial peptides

First of all, peptide or protein finally?
It's very important question.
After that your gel didn't destaine very well and you didn't load ladder(marker) thus it's not possible answer you clearly.
My part of work is isolation and characterization of antimicrobial peptides but for that first I need to extract protein from sample and for that I m not getting clear bands I m not yet used any marker here
You have to load a marker for checking your gel.
When you load total protein is typical you got smear.
