In evaluating commercially available kits for exosome isolation, the main question is: can I consider wound fluids/ascites' composition/characteristics similar to serum? I mean, for those kits which distinguish between plasma and serum, where can I place my wound fluids? Is the presence/absence of clotting factors the major determinant?
I think that if you use a different fluid than one used for the development of a kit, the efficiency would be just diferent. Don't know if better of worse, that woul depend on the kit. For instance, exoQuick is only a web made of polymers (originaly used for virion concentration) that thraps every small particles inclluding exosomes. You can asumme that a less visqueous fluid would result in less contaminants in your final pellet... or more particles would flush with the fluid during centrifugation..... don't know if I helped !!
Wound fluid composition is influenced by the state of the wound and the phase of healing and therefore the extracted fluid will be more or less rich in clotting factors. Same considerations should be made for the composition of ascitic fluid.
Both fluids isolated, in different stages/phases of wound healing and/or disease, contain markedly higher percentage of proteins compared to that of the healthy plasma/ serum. This clearly, as Christophe mentions, influences the viscosity and purity of your sample and consequently the quality of isolated exosome fraction.
Well, a careful consideration of the composition of the starting sample, will most probably guide you to the most effective exosome-isolation kit.

good question. there are 5 Total exosome isolation reagents (for misc fluids and CM) but they dont cover all body fluids. I'd recommend to try the reagent for "other body fluids" and test 2 "extreme" protocols- one for the fluids rich in exosomes, another one for poor. see which one woks best for you
