I've to estimate the chlorophyll content in fresh leaf of camellia sinensis sp. So, I collected fresh leaf and dried at 50°C for 7 hour and preserved in dark conditions. Due to some inconveniency I've to carry out my analysis after a week. Now, will there any significant change in chlorophyll content due to a week preservation?

Oven drying reduces chlorophyll content compared to room or freeze dried samples. So even though you waited a week, chloropohyll would already be reduced due to the original drying process. Perhaps redo the experiment by room drying the samples :)

Why oven drying? A better estimate is obtained when you use fresh samples. If you have to store the samples, it has to be in refrigerator.
Hi, Oyetunji. I went for oven dry to reduce moisture and to grind the leaf to powder. Fresh leaf chopped and chlorophyll extract by using acetone will be the best procedure? What's your conception in this regard? Please.
You should do it with fresh leaves. I did it using N,N-Dimethylformamide. (see Page 94)
Always better if go with fresh, otherwise from frozen samples stored at -80 degree C (obtained from intact plants and immediately placed in liquid N then shifted into deep freezer).
The chlorophyll content of dried sample and fresh leaves varies. I worked on both kinds of samples and checked the content using the spectrophometric method using acetone washings.
Fresh leaf is the best source, I found in my experiment also. However, thanks to all for your informative suggestion.
