can anyone give suggestion of how to degrade PLGA scaffold in order to do qPCR?
and also how to visualize the cells in the scaffold by microscopy because the scaffold is non transparent?
Hi I dont know about the degradation
But one suggestion to visualise the cells is to tag them using fluorescent signal using CELL SIGNAL KIT(Before putting them in th scaffold)
Of course this is not good if you care about the histology , but it just show you cell viability and distribution.
Or you might try normal tissue staining with Hematoxylin & Eosin to stain cell nucleous different from the extra-cellular matrix
yes but the microscope can't pass the structure,my EScells express beta gal gene so I can reveal them with Xgal or with FITC Ab anti-lacZ so the main problem is the fact that I can visualized the cells on or in the scaffold.
