You will probably want to try a few different concentrations of doxycycline to achieve an acceptable expression level of your inducible gene. I'd recommend trying a range from 1 – 1000 ng/ml. You should be able to detect transgene expression within 24 hours, but you may see a stronger effect at 48 or 72 hours depending on your system (e.g shRNA).
