Hello I have a question about doxorubicin hydrochloride ? has doxorubicin in own molecule net positive charge or not? I mean when we dissolve doxorubicin hydrochloride in distilled water doxorubicin has positive charge or not?
it is said that "Dox-HCl salt is dissolved in water it dissociates into doxorubicin, H+, and Cl- "
I want to know about net charge of doxorubicin molecule in water ?

Sounds that DOX(HCL) has the 0 formal charge which is calculated by PubChem.

Besides, personally, i think the structural charge distribution in DW is also another prove that this claim makes quitely physical Sense.

Yes I know that the Dox-HCl net charge is 0, I asked about its charge after dissolving in DW, it means DOX after dissolving has net charge or not ? if yes? positive or negative and why?

H+ Will interact with free amino group of the own molecule (i mean the DOXO itself)
Then the net charge of molecule change to positive
But this cation Then will be surrounded by Cl_ Negative ion to acheieve an ioniq soluable salt (ioniq liquid) with a certain Kd constant (an equilibrium)
((So eventually as it dissolves in DW
It is UNCHARGED (because of sorounding by Cl_)
but by changing pH it will also do another trend))

To tell the answer based on my knowledge, first we should know what is the meaning of salt , and how salt of this drug is formed. Salt is the product of interaction between acid and a base , here the paren drug (doxorubicin) is a weak base .and HCl is acid. But here interaction is between cation of base and anion of acid will form a salt .now we should know what is the cation of the base (doxorubicin)here: very interesting point is that the cation of base(doxorubicin)is obtained by protonation of N of the doxorubicin( via dative bond) and form of NH3+ . and the anion of acid is Cl- as the result this 2( +&-) charge make a ionic bond and the salt is formed (without releasing water) . Let’s come to your question ma’am . Yes HCl will convert to H+&Cl- but!!! The H+ will go to N of Doxorubicin and make it protonated means . This is a salt formations without producing water molecule, doxorubicin formula is C27H29NO11 & doxorubicin HCl formula is C27H30NClO11 . The salt can be dissolve in water and form C27H30O11N+ and Cl- . It’s anion and cation . To add a point salt is in solid state (s) once it’s dissolved in water it coverts to (aq). The last point : protonated doxorubicin will not be surrounded with Cl- but both cation and anion formed will be surrounded with water molecules, by hydration reaction this is how it dissolve in water only ! if its surrounded with Cl- it will form salt again and precipitation will form, Khatereh Khorsandi
