My question is how I can increase the quantity of RNA isolated from cells in wound edge in this case from keratinocytes? I have done the experiment and still the quantity of isolated RNA is low for RNA seq. does anyone have experience who could help me?

Unfortunately, the only way is increasing your sample. If you are working with isolated cells, try to get the maximum number that your kit or your extraction protocol requires.
If you are working with tissue, 50mg of tissue is the usual amount of tissue for Trizol or other phenol-chloroform protocols.
Another issue is your tissue may not express a lot of RNA. Try to access how much RNA you can get from keratinocytes in other studies, but for me they did not express a lot of RNA, like other cell type with higher expression activity like hepatocytes.
